    Ac4GalNAlk (Tetraacetylated N-(4-pentynoyl)galactosamine) is a synthetic, chemically modified sugar derivative primarily used as a Metabolic Chemical Reporter (MCR). Its fundamental function is to track, visualize, and map protein glycosylation (specifically O-linked glycoproteins) inside living cells and tissues through a technique known as Metabolic Oligosaccharide Engineering (MOE). 

    How Ac4GalNAlk Functions (Step-by-Step)
    • Cellular Entry (The Ac4 Advantage): The four acetyl groups (Ac₄) mask the negative charges of the sugar. This makes the molecule highly hydrophobic, allowing it to easily diffuse across the fatty cell membrane.
    • Intracellular Activation: Once inside the cell, non-specific enzymes called carboxyesterases strip away the acetyl groups, trapping the modified sugar inside the cytoplasm.
    • Hijacking the Salvage Pathway: The cell mistakes the remaining GalNAlk monomer for natural N-acetylgalactosamine (GalNAc). Cellular enzymes (such as the GALK2 kinase and AGX1/2 pyrophosphorylases) process it into an active nucleotide-sugar (UDP-GalNAlk).
