1-(2-CHLORO-PYRIDIN-4-YL)-ETHANONE Use and Manufacturing
Step 2: To a stirred solution of 2-chloro-N-methoxy-N-methylisonicotinamide (1.50 g 7.476 mmol) in dry THF (25 ml), was added methylmagnesiumbromide [3M in Diethyl ether; (3.8ml 12.4mmol)] drop wise at 0° C. The reaction mixture was stirred at 0° C for 2hours. The progress of the reaction was monitored by TLC (2:8, Ethyl acetate: pet ether). The reaction mixture was quenched with saturated ammonium chloride solution and then extracted with ethyl acetate (2 x 75 ml). The combined organic layer was washed with water (100 ml) followed by brine (50 ml) and then dried over anhydrous NaMethylmagnesium bromide solution in THF (3 M, 100 mL, 300 mmol) was dropwisely added to a flask containing a solution of 2-chloroisonicotinonitrile (21 g, 151.6 mmol) in THF (300 mL) at 0 °C under nitrogen and the mixture was stirred for 24 h at rt. Then the mixture was poured into a mixture of concentrated hydrochloride (25 mL) and ice (400 g) and extracted with EtOAc (2x400 mL). The organic layers were washed with brine, dried and concentrated under reduced pressure. The residue was purified via Combi-Flash on silica gel (15 percent petroleum ether/EtOAc) to afford l-(2-chloropyridin-4-yl)ethanone (7.5 g, 32 percent) as a white solid. MS ESI calc'd for C
