H-D-VAL-NH2 HCL Use and Manufacturing

325 ml of water was added to 25.0 g of A solution of 3-bromo-5-tluoropicolinonitriie (300 mg, 1.49 mmol), A solution of 2-(1H-benzo[d][1, 2, 3]triazol-1-yloxy)-4-(4-methylthiophen-2-ylamino)pyrimidin-5-carboxamide (70 mg, 0.19 mmol), Commercially available B5.1 (1.02 g, 5.86 mmol) was dissolved in -30 mL DMF with 1.5 mL DIEA. To this stirring solution was added 850 mg (5.86 mmol, 1 eq) of 7- aminoquinoline. The reaction was stirred at room temperature for 30 minutes. An aliquot (~.5 mmol) was taken aside, and to it was added 0.1 mL DIEA and H-D-Val-NH2 HCl. This reaction mixture was stirred for 3 hours at 50C and then cooled. Water and DCM was added and the layers were separated. The organic layer was washed with 10% NaHC03 and was concentrated. Crude B5.2 was dissolved in -10 mL MeOH. To this solution, -50 mg K2C03 and -1 mL H202 (40% by wt) were added. The reaction was stirred at 50C for 30 minutes and then was concentrated. The crude was purified by rpHPLC to give the title compound. MS found for C19H21N702 as (M+H)+380.2.General procedure: Step 5: Compound B1.4 was dissolved in ~3 mL NMP. To it was added ~2eq mCPBA. The reaction mixture was stirred at RT for 45 minutes. To it then were added commercially available (R)-2-aminobutanamide HCl and DIEA. The mixture was stirred for 90 minutes at 120C bath. This mixture was then subjected to preparative HPLC to isolate the title compound. MS found for C16Hi8N802 as (M+H)+ 355.3.Step 1: To a mixture of 3, 5-dichloropyrazine-2-carbonitrile (500 mg, 2.84 mmol) and
