Mavacamten.

Mavacamten (MYK-461, SAR439152) is a small molecule modulator of cardiac myosin (cardiacmyosin) and regulates nodal hypertonic contraction in hypertrophic cardiomyopathy. In vitro studies have shown that MYK-461 reduces steady-state ATPase activity by inhibiting the rate of phosphate release from β-cardiac myosin-S1.MYK-461 regulates multiple steps in the chemomechanical cycle of myosin: in addition to decreasing the rate-limiting step in the cycle (phosphate release), MYK-461 reduces the number of myosin-S1 heads (during the transition from weakly-bound to strongly-bound state). MYK-461 also reduces the number of myosin-S1 heads (during the transition from the weakly bound to the strongly bound state, myosin-S1 heads can interact with actin filaments). myosin-S1 also reduces the rate at which myosin binds to ADP-actin, as well as the rate at which ADP is released from myosin-S1.
