Zinc oxide (ZnO) is an inorganic compound widely used in pharmaceuticals, cosmetics, and medical devices. It exists as a white, odorless powder with mild astringent and protective properties. In medicine, it is commonly applied topically in creams, ointments, and pastes (e.g., diaper rash treatments, sunscreens) and is also used in oral formulations (e.g., dietary supplements, wound-healing agents). Its versatility stems from its antimicrobial, anti-inflammatory, and UV-blocking effects.
Mechanism of Action
Zinc oxide exerts multiple therapeutic actions:
Topically : Forms a protective barrier on the skin, absorbs moisture, and soothes irritation. It releases zinc ions, which exhibit antimicrobial activity and promote wound healing by enhancing collagen synthesis and reducing inflammation.
In sunscreens : Acts as a physical UV filter, scattering and reflecting UVA/UVB rays.
Orally : Supports immune function, enzyme activity, and tissue repair as an essential trace mineral.
Dosage and Administration
Topical use (e.g., diaper rash, minor wounds) : Apply a thin layer to affected areas 1–3 times daily.
Sunscreens : Use as directed (typically 5–25% ZnO concentration).
Oral supplements : Adults typically take 15–40 mg/day (elemental zinc); higher doses for deficiencies require medical supervision.
Note : Oral zinc oxide is less common; zinc salts (e.g., zinc gluconate) are preferred for supplementation.
Adverse Reactions
Topical : Rare hypersensitivity (itching, redness); prolonged use on broken skin may cause dryness.
Oral : Nausea, vomiting, metallic taste, or diarrhea (especially at high doses >40 mg/day). Chronic excess intake can lead to copper deficiency and neurological effects.
