Rolapitant was approved by the U.S. Food and Drug Administration (FDA) in September 2015. It is the latest long-acting, highly selective NK-1 receptor antagonist. Rolapitant has a long half-life of up to 180 hours and, compared to other NK-1 receptor antagonists, including aprepitant, offers more significant advantages, such as selectively binding to human NK-1 receptors and having high affinity according to Chemicalbook. Additionally, unlike other NK-1 receptor antagonists (such as aprepitant and netupitant), rolapitant does not inhibit or induce CYP3A4.
