    1. Description

    4-Butylresorcinol is a synthetic resorcinol derivative belonging to the class of alkylresorcinol compounds. It appears as a white to off-white crystalline powder or a pale yellow powder, with the molecular formula C₁₀H₁₄O₂ and a molecular weight of approximately 166.22 Da. Its melting point is 49–53°C, and its boiling point is approximately 324.3°C. This compound is highly soluble in ethanol, dimethyl sulfoxide, and oily solvents, and slightly soluble in water (solubility in water is 0.6 g/L at 25°C). It should be stored away from light, in a sealed container, in a cool, dry place, at 2–8°C or room temperature. As a highly effective tyrosinase inhibitor, 4-butylresorcinol enhances its lipophilicity and enzyme-binding capacity through the butyl substitution on the resorcinol ring, enabling it to penetrate the skin more effectively and inhibit melanin production. This compound is hailed as the “gold standard for skin whitening” in the skincare industry and is widely used in Asian countries such as South Korea and Japan.

    2. Application

    (1) Cosmetics and Skin Brightening: As a functional brightening ingredient, it is widely incorporated into skincare products such as serums, creams, lotions, face masks, and spot-fading pens to improve dull and uneven skin tone. In South Korea, the maximum permitted concentration is 0.1%; in Japan, it is 0.3%. It has been approved for use in cosmetics in both the European Union and China.

    (2) Spot-Fading Products: Used to lighten skin pigmentation issues such as melasma, freckles, age spots, sunspots, and post-inflammatory hyperpigmentation. Clinical studies indicate that 4-butylresorcinol at concentrations of 0.1%–0.5% has a significant improving effect on melasma.

    (3) Skin-brightening skincare products: Added to skin-brightening product lines to brighten skin tone, improve skin texture, and enhance skin radiance. Its skin-lightening efficacy surpasses that of arbutin and kojic acid, making it one of the most potent tyrosinase inhibitors currently available.

    (4) Skin Care Research: Used as a standard or tool compound in studies on skin whitening mechanisms, serving as a tyrosinase inhibitor to evaluate the regulation of melanin synthesis.

    (5) Synergistic Whitening Formulations: Often combined with other whitening ingredients such as niacinamide, vitamin C derivatives, kojic acid, and azelaic acid to exert synergistic effects through multi-target mechanisms of action.

    3. Main Efficacy

    (1) Potent inhibition of tyrosinase activity: By competitively inhibiting tyrosinase activity, it blocks the key step in the conversion of tyrosine to dopa and dopaquinone, effectively reducing melanin biosynthesis. Its inhibitory activity is significantly stronger than that of kojic acid, arbutin, and hydroquinone, with a half-maximal inhibitory concentration (IC₅₀) for tyrosinase as low as the nanomolar range. Molecular docking studies indicate that 4-butylresorcinol forms hydrophobic interactions with the tyrosinase protein via its butyl chain, while its phenolic hydroxyl group forms a coordinate bond with the copper ion at the active site.

    (2) Significantly reduces melanin production: In melanocytes (B16F10 mouse melanoma cells and normal human melanocytes) and 3D skin models, it concentration-dependently reduces melanin content, lightens existing pigmentation, and prevents new pigment formation. At a concentration of 20 μg/mL, 4-butylresorcinol inhibits melanin synthesis by over 80%.

    (3) Lightens various types of pigmentation: It has a significant lightening effect on melasma, freckles, age spots, sunspots, and post-inflammatory hyperpigmentation, improving skin pigmentation abnormalities. In clinical trials, after 8 weeks of use at a 0.3% concentration, the area and severity index score of melasma decreased by approximately 40%–50%.

    (4) Brightening and evening out skin tone: Continuous use results in a more even and luminous complexion, improving dullness, sallowness, and uneven skin tone while enhancing overall skin brightness. Studies show that skin brightness (L* value) significantly increases after 4 weeks of use.

    (5) High Skin Penetration: The butyl substitution enhances the molecule’s lipophilicity (logP approximately 3.2), enabling it to effectively penetrate the skin’s stratum corneum and reach melanocytes in the basal layer to exert its effects. In vitro skin penetration tests show that 4-butylresorcinol has a skin penetration rate approximately 5–10 times higher than that of arbutin.

    (6) Stability and Safety: It possesses high chemical stability and is unlikely to oxidize or discolor in formulations. At recommended concentrations (≤0.3%), it exhibits low skin irritation and is suitable for short-term intensive brightening use. However, compared to traditional brightening agents, its irritation potential is slightly higher than that of arbutin and niacinamide; therefore, it is recommended to start with low concentrations.

    (7) Antioxidant Support: It possesses some free radical scavenging capacity, helping to reduce oxidative stress damage to skin cells and protect the skin by reducing UV-induced reactive oxygen species (ROS) production.

    (8) Anti-inflammatory Activity: Studies indicate that 4-butylresorcinol also exhibits anti-inflammatory effects, inhibiting lipopolysaccharide-induced prostaglandin E2 and nitric oxide production, and alleviating UV-induced skin inflammatory responses.

    (9) Synergistic Skin-Brightening Effects: When combined with other skin-brightening ingredients such as niacinamide, vitamin C derivatives, kojic acid, and azelaic acid, it produces synergistic effects by inhibiting different stages of melanin synthesis (including tyrosinase activity, melanin transfer, and melanocyte proliferation), thereby enhancing the overall skin-brightening effect.
