2-Amino-3-chlorobenzaldehyde Use and Manufacturing

General procedure: To a solution of 1a (1.2 g, 7.61 mmol) in CHAdding urea (10.45g, 174.05mmol) in a stainless steel sealed tube with 2-amino-3-chlorophenyl carbaldehyde (1.80g, 11.60mmol), The reaction temperature was raised to 180 deg.] C for 1.5 hours. The reaction was stopped, cooled to room temperature, water (50 mL), filtered, washed with acetone (10mL) and ethyl acetate (10 mL) slurried, filtered, and dried in vacuo to obtain a cake. The filter cake was added to phosphorus oxychloride (50 mL) was refluxed for 3 hours. The reaction was stopped, cooled to room temperature, ice water (50 mL) quenched with solid sodium bicarbonate and sodium hydroxide to adjust the pH 9; (20mLx3) and extracted with dichloromethane and the combined organic phases, the organic phase was washed with saturated brine water (20mLx3), dried over anhydrous sodium sulfate, filtered, and the filtrate was concentrated under reduced pressure and purified by column chromatography (petroleum ether / ethyl acetate (v / v) = 20/1) to give the title compound as a pale yellow solid (973 mg, 42.3%).
