One of the key highlights of Yasheng's clinical research is the novel multikinase inhibitor Olverembatinib, which has demonstrated remarkable antitumor activity in succinate dehydrogenase (SDH)-deficient neoplasms. The results of preclinical studies show that Olverembatinib outperforms other tyrosine kinase inhibitors in tumor cells with SDH deficiencies. Mechanistic studies have revealed that Olverembatinib exerts its antitumor effects by modulating multiple signaling pathways associated with SDH-deficient tumors, such as hypoxia, angiogenesis, cell proliferation, and apoptosis. These findings provide a solid scientific foundation for the potential clinical application of Olverembatinib in this challenging subset of tumors.
Synergistic Inhibition of Tumor Growth in Prostate Cancer Models:
Ascentage Pharma' research also highlights the synergistic effects of the embryonic ectoderm development (EED) inhibitor APG-5918 and the MDM2 inhibitor alrizomadlin in preclinical models of prostate cancer (PCa). Through their regulation of DNA methylation, cell cycle progression, and apoptosis-related pathways, the combination of APG-5918 and APG-115 has demonstrated enhanced antitumor activity. These findings lay the groundwork for the clinical development of a combination therapy involving APG-5918 and APG-115 in the treatment of prostate cancer.
APG-2449 Enhances Antitumor Efficacy in Epithelial Ovarian Cancer:
Ascentage Pharma' research has also revealed the potential of APG-2449, a novel focal adhesion kinase (FAK) inhibitor, in the treatment of epithelial ovarian cancer (EOC). APG-2449 has been shown to inhibit tumor cell migration and synergistically enhance the antitumor efficacy of PEGylated liposome doxorubicin (PLD) in EOC cells. These findings provide a promising direction for the further clinical development of APG-2449 in combination with PLD for the treatment of ovarian cancer.
