Semaglutide: The Dose Is Increased From 1mg To 2mg, The Efficacy Is Further Enhanced
On March 28, 2022, Novo Nordisk announced that the FDA approved a new dose of semaglutide for the treatment of type 2 diabetes: 2 mg. The initial dose of semaglutide was 0.5 mg, and after the supplementary phase III clinical trials, the dose was increased to 1 mg and 2 mg successively.

According to the phase III SUSTAIN FORTE data, 68% of patients achieved HbAc1 <7% in the 2mg dose of semaglutide for 40 weeks, compared with 58% in the 1mg dose group.

The hypoglycemic and weight loss effects of the semaglutide 2 mg dose group were better than those of the 1 mg dose group.

Summarize
Dose adjustment is an important trend in the competition for the development of GLP-1 receptor agonists. The initial dose of Lilly dulaglutide was 1.5 mg, which was later increased to 3.0 mg and 4.5 mg. Semaglutide was increased from 0.5mg to 1mg and 2mg. Eli Lilly's GLP-1R/GIPR dual-target agonist Tirzepatide selected the doses of 5mg, 10mg, and 15mg in the third phase of the clinical trial, and the dose of semaglutide in the control group was 1mg.
