Function and Efficacy

1. Pharmacology Ranitidine has the effect of competitively blocking the binding of histamine to H2 receptors. It inhibits gastric acid action, which is 5 to 12 times that of cimetidine in terms of moles. Therefore, it is a potent H2 receptor blocker; 2. Toxicity The oral LD50 of mice is 1440~1750mg/Kg. The maximum non-toxic dose per day for 5 consecutive weeks is 500mg/Kg for rats (male), 250mg/Kg for rats (female), and 40mg/Kg for dogs. The maximum non-toxic dose per day for 26 consecutive weeks is 100mg/Kg for rats and 40mg/Kg for dogs. Mice were orally administered 100~200mg/Kg for 114 weeks, and rats were orally administered 100~2000mg/Kg for 129 weeks, and no carcinogenic effect was observed.

Appearance

This product is a sugar-coated tablet or a film-coated tablet, which appears off-white or slightly yellow after removing the coating.

Indication

Used to treat duodenal ulcer, gastric ulcer, reflux esophagitis, Zollinger-Ellison syndrome and other high gastric acid secretion diseases.

Usage and Dosage

1. Oral administration, 150 mg (1 tablet at a time), twice a day, or 300 mg (2 tablets at a time) once before bedtime; 2. Maintenance treatment: oral administration, 150 mg (1 tablet at a time), once every night; 3. For patients with severe renal disease, the half-life of ranitidine is prolonged, and the dose should be reduced to 75 mg (half a tablet at a time), twice a day; 4. For the treatment of Zollinger-Ellison syndrome, a large amount should be used, 600~1200 mg per day (4~8 tablets per day).

Adverse Reactions

1. Common symptoms include nausea, rash, constipation, fatigue, headache, dizziness, etc.; 2. Compared with cimetidine, the adverse reactions of damaging kidney function, gonadal function and central nervous system are milder; 3. A small number of patients suffered mild liver damage after taking the drug, and the symptoms disappeared after stopping the drug, and the liver function returned to normal. It was suspected that it might be a drug allergic reaction and had nothing to do with the dosage of the drug; 4. Long-term use can continuously reduce the acidity of gastric juice, which is conducive to the reproduction of bacteria in the stomach, thereby reducing nitrates in food to nitrites and forming N-nitroso compounds.

Precautions

Pregnant and lactating women are prohibited from using this product. Children under 8 years old are prohibited from using this product.

Drug Interactions

1. When used in combination with warfarin, lidocaine, diazepam, propranolol and other drugs that are metabolized by the liver, the blood concentration of ranitidine will not increase and cause toxic and side effects; 2. When used in combination with anticoagulants and anti-epileptic drugs, it is safer than cimetidine; 3. When used in combination with procainamide, the clearance rate of procainamide can be reduced; 4. It can reduce liver blood flow, and therefore when used in combination with propranolol, lidocaine and other drugs whose metabolism is greatly affected by liver blood flow, it can delay the effects of these drugs.
