1. Description

Saxifragin is a naturally occurring isocoumarin compound primarily derived from plants in the Saxifragaceae family (such as Saxifraga sarmentosa and Saxifraga sericea), though it is also found in various other plants, including Coptis chinensis. It appears as white, loose, needle-like crystals or a crystalline powder; it is odorless and has a bitter taste; it is prone to discoloration when exposed to light or heat. It is soluble in methanol and slightly soluble in water or ethanol; its melting point ranges from 232 to 240°C. It possesses various biological activities, including antitussive, expectorant, anti-inflammatory, antioxidant, hepatoprotective, antitumor, and neuroprotective effects.

2. Application

(1) Research on Respiratory Diseases: Used in clinical studies of respiratory diseases such as chronic bronchitis, bronchial asthma, and emphysema, where it exerts antitussive and expectorant effects to alleviate symptoms such as coughing and sputum production.

(2) Anti-inflammatory and Immunomodulatory Research: Used in studies of inflammatory diseases such as ulcerative colitis, idiopathic pulmonary fibrosis, atherosclerosis, and severe sepsis. Research indicates that rock cabbage extract can reduce inflammatory responses by inhibiting the TLR4/NF-κB/NLRP3 inflammatory signaling pathway.

(3) Antioxidant and Cardiovascular Protection Research: Used to study free radical scavenging, inhibition of lipid peroxidation, and protection of DNA from oxidative damage, exerting a protective effect in cardiovascular disease models such as atherosclerosis and ischemia-reperfusion injury.

3. Main Efficacy

(1) Antitussive and expectorant effects: Used to treat chronic bronchitis, it exerts antitussive, expectorant, and bronchodilatory effects, making it a commonly used clinical medication for relieving cough and clearing phlegm.

(2) Anti-inflammatory effects: By inhibiting the NF-κB/NLRP3 signaling pathway, it reduces the production of pro-inflammatory cytokines such as interleukin-1β and tumor necrosis factor-α, thereby alleviating inflammatory responses. In ulcerative colitis models, it can improve intestinal inflammation by inhibiting the TLR4/NF-κB pathway. In idiopathic pulmonary interstitial fibrosis models, it exerts therapeutic effects by inhibiting the NF-κB/NLRP3 pathway.

(3) Antioxidant Effects: It scavenges superoxide anion radicals, inhibits lipid peroxidation, and protects DNA from oxidative damage caused by hydroxyl radicals. In vitro experiments show that its antioxidant activity surpasses that of the classic antioxidant vitamin C.

(4) Hepatoprotective effects: By inhibiting inflammation mediated through the IL-6/JAK2/STAT3 signaling pathway and exerting antioxidant effects, it exerts a protective effect against carbon tetrachloride-induced acute liver injury. In a non-alcoholic fatty liver disease model, it reduces inflammatory responses and oxidative stress, regulates lipid metabolism, and improves hepatic steatosis and necrosis.

(5) Antitumor Effects: It inhibits the proliferation of malignant glioma cells, a mechanism associated with reduced phosphorylation of the PI3K-AKT-mTOR signaling pathway. In radiation-resistant oral squamous cell carcinoma cells, it restores radiosensitivity by stabilizing PTEN protein, inhibiting AKT phosphorylation, and suppressing HK2 expression. In non-small cell lung cancer cells, it inhibits cell proliferation and promotes apoptosis by suppressing the NF-κB pathway.

(6) Neuroprotective effects: In a mouse model of cerebral ischemia-reperfusion, it exerts neuroprotective effects by scavenging free radicals and inhibiting lipid peroxidation.

(7) Anti-hyperuricemic activity: As a xanthine oxidase inhibitor, it has the potential to lower uric acid levels.

(8) Antimicrobial and antiviral activity: It exhibits broad-spectrum antimicrobial activity, inhibiting Shigella, Staphylococcus aureus, Pseudomonas aeruginosa, and others, and also possesses antiviral activity.
