    1. Description

    Ecdysone is a naturally occurring steroid hormone belonging to the ecdysone family. It serves as a key regulatory hormone in the molting and metamorphosis processes of insects and other arthropods. It appears as a white to off-white solid powder, soluble in acetic acid, ethanol, methanol, or dimethyl sulfoxide; slightly soluble in chloroform; and insoluble in water. Its molecular formula is C₂₇H₄₄O₆, and its molecular weight is approximately 464.6. It should be stored at -20°C in a light-protected, sealed container. This compound is a precursor to 20-hydroxyecdysone, the primary ecdysone in insects, and exerts its effects through heterodimeric receptor complexes to regulate the expression of genes associated with molting and metamorphosis.

    2. Application

    (1) Psoriasis Research: Studies have shown that Licoisoflavone B regulates the reprogramming of keratinocyte lipid metabolism by targeting stearoyl-CoA desaturase 1, thereby inhibiting IL-17-induced excessive keratinocyte proliferation. In an imiquimod-induced mouse model of psoriasis, oral administration of Licoisoflavone B (40–160 mg/kg) significantly reduced epidermal hyperplasia and inflammatory responses.

    (2) Drug Metabolism and Interaction Studies: As a selective inhibitor of cytochrome P450 enzymes, it is used to study drug interactions and metabolic pathways. Licoisoflavone B exhibits reversible competitive inhibition of CYP2C8 (IC₅₀ = 7.4 μM, inhibition constant Ki = 7.0 μM) and reversible mixed inhibition of CYP2C9 (IC₅₀ = 4.9 μM, Ki of 1.2 μM), and exerts moderate inhibition of CYP2B6 via a dual mechanism of irreversible and reversible inhibition (IC₅₀ of 16.0 μM).

    (3) Oxidative stress studies: Investigations into the mechanism of inhibiting lipid peroxidation. In a copper-induced model of protein oxidation modification in mouse brain homogenate, Licoisoflavone B demonstrated inhibitory effects on protein oxidation modification, with its activity attributed to the compound’s hydroxyl groups and benzopyran structure.

    (4) Antiviral Studies: Based on molecular docking and dynamics simulations, Licoisoflavone B exhibits strong binding affinity for the nucleoprotein of Lassa virus, maintains structural stability in 100 ns molecular dynamics simulations, and demonstrates favorable electronic properties as indicated by density functional theory analysis, suggesting its potential as a therapeutic candidate for Lassa fever.

    (5) Phytochemical and Quality Control Studies: As a characteristic component of plants such as licorice, it is used in the quality control of traditional Chinese medicinal materials, fingerprint analysis, and the screening of bioactive compounds.

    (6) Antifungal Studies: It is used in studies to inhibit the growth of hyphae in arbuscular mycorrhizal fungi, exploring its potential role in plant-fungus interactions.

    3. Main Efficacy

    (1) Anti-psoriatic effects: By targeting stearoyl-CoA desaturase 1 to regulate lipid metabolic reprogramming, it inhibits the IL-17-induced upregulation of stearoyl-CoA desaturase 1 and lipid droplet accumulation in keratinocytes; it also suppresses the expression of keratin 17 and Ki67, markers of excessive keratinocyte proliferation; In a murine model of imiquimod-induced psoriasis, it reduces Th17 cell differentiation and interleukin-17 production, exerting dual antiproliferative and immunomodulatory effects.

    (2) Inhibition of cytochrome P450 enzyme activity: It inhibits human CYP2C8, CYP2C9, and CYP2B6. The strongest inhibitory effect is observed for CYP2C9 (IC₅₀ = 4.9 μM), exhibiting a mixed inhibition mechanism (Ki = 1.2 μM); It exhibits competitive inhibition of CYP2C8 (IC₅₀ = 7.4 μM, Ki = 7.0 μM); for CYP2B6, it produces moderate inhibition through a dual mechanism of irreversible and reversible inhibition (IC₅₀ = 16.0 μM, time-dependent inhibition of 34.8%, with 76.4% residual activity after washing). It also exhibits weak inhibition of CYP3A4 (IC₅₀ = 26.7 μM).

    (3) Inhibition of lipid peroxidation: It effectively inhibits lipid peroxidation reactions with a half-maximal inhibitory concentration (IC₅₀) of 2.7 μM, protecting cell membranes and proteins from oxidative damage. Its inhibitory activity is associated with the compound’s hydroxyl groups and benzopyran structure.

    (4) Potential antiviral activity: Computational modeling studies indicate that Licoisoflavone B exhibits strong binding affinity for the Lassa virus nucleoprotein, maintaining conformational stability in 100 ns molecular dynamics simulations. Principal component analysis further confirms this conformational stability, while density functional theory analysis reveals favorable electronic properties, and pharmacokinetic and toxicity predictions show excellent characteristics.

    (5) Antifungal activity: At a concentration of 0.63 μg/plate, it strongly inhibits the growth of hyphae in arbuscular mycorrhizal fungi.
