      Usage Method  


      • 1. Application Scope & Preconditions


        This product is a professional research-grade mitochondrial-derived peptide material, only applicable to laboratory scientific research, in vitro cell experiments, exploration of mitochondrial functional mechanisms, metabolic pathway analysis and other basic academic research. It is prohibited for clinical treatment, human trial, food addition, cosmetic formulation and any other non-research industrial production activities. All operators shall possess basic experimental qualifications and be familiar with laboratory safety regulations. Before operation, please read the product description and compliance statement thoroughly to clarify the product nature and scope of application.

        2. Pre-experiment Preparation


        2.1 Preparation of Environment & Apparatus


        1. All operations must be conducted in a laboratory that meets cleanliness standards. A biosafety cabinet or clean bench is recommended. Turn on relevant equipment 30 minutes in advance to purify the operating space and prevent microbial and particulate contamination of samples.
        2. Prepare sterile laboratory tools including sealed vials, pipettes, sterile tips, centrifuge tubes, glass rods, sealing films, alcohol swabs, disposable sterile gloves, dust-proof masks and goggles. All utensils shall be sterilized and dried before use.

        2. Sample tempering: Take the vial of MOTS-c lyophilized powder out from -20°C storage and place it at room temperature for 15 to 20 minutes to balance the internal temperature, so as to avoid excessive foam and powder adhesion during dissolution due to temperature difference.
        3. Disinfection: Wipe the rubber stopper of the vial, pipette interfaces and tube openings with alcohol swabs, and wait until fully dry before use. Maintain strict aseptic operation throughout the whole process.

        3. Standard Dissolution Procedures


        1. Concentration calculation: Calculate the required solvent volume according to experimental design. For reference: dissolve 5mg powder in 0.5mL solvent, 10mg powder in 1mL solvent and 20mg powder in 2mL solvent to prepare the solution at the concentration of 10mg/mL. The concentration can be adjusted as required.
        2. Solvent addition: Draw the preheated solvent with a sterile pipette and add it slowly along the inner wall of the vial. Do not impact the powder directly to prevent excessive foam and structural damage of peptide molecules.

        4. Application Methods for Different Research Scenarios


        4.1 In Vitro Cell Experiments


        1. Cell pre-cultivation: Culture target cells until they reach the logarithmic growth phase with confluence of 70% to 80%, then replace with fresh culture medium for standby use.

        4.2 Mitochondrial Function Research


        1. Mitochondria extraction: Isolate complete mitochondria from fresh tissues or cells via differential centrifugation. All operations shall be performed at 4°C to maintain the integrity and activity of mitochondria.
        2. Reaction system preparation: Add appropriate amount of MOTS-c solution to the mitochondrial reaction buffer, with final concentration ranging from 1μM to 5μM. Set up blank control groups and negative control groups to eliminate interference factors.
        3. Functional detection: Detect the activity of mitochondrial respiratory chain, ATP production, reactive oxygen species level and calcium homeostasis by using fluorescence spectrophotometer, chemiluminescence detector and other devices, so as to analyze the impact of MOTS-c on physiological functions of mitochondria.

        4.3 Research on Metabolic Mechanisms & Molecular Pathways


        1. Sample treatment: Extract total protein, total RNA or metabolites from cells and tissues after intervention with MOTS-c for subsequent molecular biological tests.
        2. Index detection: Detect the mRNA expression level of metabolism-related genes via qRT-PCR, test protein expression and phosphorylation level by Western Blot, and analyze the content changes of carbohydrates, lipids, amino acids and other metabolites through metabolomics technology, so as to explore the molecular mechanism of MOTS-c in metabolic regulation.

        5. Post-use Treatment & Notes


        5.1 Storage of Residual Solution


        1. Seal the remaining stock solution and sub-packaged solution tightly immediately after use. Store them at 2°C to 8°C away from light, and use up within one month to avoid activity decline and contamination.
        2. Repeated freezing and thawing of diluted working solution are prohibited. Prepare the solution according to actual experimental dosage every time, and do not reuse the leftover working solution to avoid experimental errors.

        5.2 Safety Protection & Waste Disposal


        1. Wear gloves, goggles and masks during the whole operation. Avoid direct contact of skin and mucous membranes with powder or solution.       Storage Guidelines
