Melanotan II (MT-2) is a synthetic cyclic analog of endogenous alpha-melanocyte-stimulating hormone (α-MSH), classified as a non-selective melanocortin receptor agonist. It is predominantly applied in preclinical research involving cutaneous pigment regulation, central metabolism and neuroendocrine pharmacology. With acetyl modification and cyclic peptide structural optimization, Melanotan II presents superior binding durability and proteolytic resistance compared with native α-MSH.

In vitro receptor binding assays verify that Melanotan II potently binds to four melanocortin receptor subtypes including MC1R, MC3R, MC4R and MC5R, with no agonistic activity on adrenal-specific MC2R, which means it cannot trigger aberrant cortisol production. It shows the highest affinity for MC1R expressed on epidermal melanocytes. Post binding, it initiates intracellular cAMP-PKA signaling cascades and upregulates MITF transcription, further activating rate-limiting melanogenic enzymes including tyrosinase and TRP-1 to promote eumelanin synthesis independent of UV radiation.
