Comparative Advantage of BeiGene Baiyueze® MAIC Results over Alectinib!
In recent news from BeiGene, the MAIC (Matching-Adjusted Indirect Comparison) results of Baiyueze® (Zebutinib) have showcased its advantages over alectinib in terms of progression-free survival and complete response rates. This groundbreaking analysis, based on data from the Phase III ALPINE and ASCEND trials, was presented at the 28th International Hematology Malignancies Congress held in Miami from February 29 to March 3. The findings shed light on the growing evolution of CLL (Chronic Lymphocytic Leukemia) treatments and solidify Baiyueze®'s position as a foundational therapy for this condition.
Comparing Baiyueze® and Alectinib:
The MAIC analysis compared the effectiveness of Baiyueze® and alectinib in treating relapsed or refractory (R/R) CLL patients. The results demonstrated that Baiyueze® exhibited advantages in terms of progression-free survival, complete response rates, and potential overall survival improvement. Notably, Baiyueze® emerged as the only Bruton's tyrosine kinase (BTK) inhibitor to demonstrate superior efficacy over ibrutinib in treating R/R CLL, as observed in the ALPINE trial. On the other hand, alectinib showed improvement in progression-free survival when compared to idelalisib/rituximab combination therapy in the ASCEND trial for R/R CLL patients. Additionally, alectinib demonstrated non-inferiority in progression-free survival compared to ibrutinib in the ELEVATE-RR trial for R/R CLL patients with chromosomal 17p or 11q deletion.
Significance of MAIC Analysis:
The MAIC analysis serves as a crucial tool in providing timely comparative effectiveness data to healthcare professionals. By robustly evaluating the effectiveness of Baiyueze® through the ASCEND and ALPINE trials, the analysis strengthens its foundational role in CLL treatment. Furthermore, the analysis highlights key differences in patient characteristics and the potential impact of COVID-19 on research outcomes. While MAIC generates hypotheses, it is important to conduct rigorous operations to minimize potential biases. In this study, individual patient-level data from the ALPINE trial were matched with aggregated data from the ASCEND trial using a non-anchored MAIC approach. Adjustments were made to account for the influence of the COVID-19 pandemic on the ALPINE trial results.
Implications and Future Considerations:
The MAIC results revealed that Baiyueze® achieved statistically significant improvements in progression-free survival and complete response rates compared to alectinib in both the unadjusted and baseline scenario-adjusted populations. Sensitivity analysis results were consistent with the baseline scenario results. Moreover, Baiyueze® demonstrated a sustained advantage in overall survival trends. While MAIC analysis generates hypotheses, the best way to evaluate drug safety in the absence of head-to-head data is to utilize the comprehensive existing evidence for each indication.
The MAIC results presented by BeiGene highlight the comparative advantages of Baiyueze® over alectinib in the treatment of R/R CLL patients. This analysis provides valuable insights for physicians, enabling them to make well-informed decisions regarding the effectiveness and safety of these therapies. With its global approvals in multiple indications, including the United States, China, the European Union, the United Kingdom, Canada, and Australia, Baiyueze® continues to demonstrate its potential in improving patient outcomes and solidifying its position as an essential therapy in the evolving landscape of CLL treatment.
