SN-38 (NK012) is an active metabolite of CPT-11, which inhibits DNA topoisomerase I, DNA synthesis, and causes frequent DNA single strand breaks. SN-38 can induce autophagy.
SN-38 is a bioactive metabolite of irinotecan hydrochloride (CPT-11). SN-38 causes the strongest inhibition of DNA topoisomerase I, followed by CPT, and then CPT-11. CPT-11 dose dependently transfers the position of loose DNA in the direction of the DNA incision, but SN-38 and CPT have no effect on the position of loose DNA. SN-38 inhibits DNA synthesis in a dose-dependent and time-dependent manner. The IC50 values of SN-38 are 0.077 micrometers in DNA synthesis. The inhibitory effect of SN-38 on cellular RNA synthesis is smaller than that on DNA synthesis and it does not inhibit protein synthesis. SN-38 causes frequent DNA single strand breaks in P388 cells.
After oral administration, the highest concentration of SN-38 appeared 1 hour later, and the percentage of unbound SN-38 lactone in mouse plasma was 3.4+/-0.67% at 1000 ng/mL, while the percentage of unbound at 100 ng/mL was 1.18+/-0.14%. The AUC of SN-38 lactone in nude mice containing human neuroblastoma is greater than that in non tumor animals.
